Design Notes

Enzyme encoded by thl catalyzes acetyl-CoA into acetoacetyl-CoA which is further transformed to 3-hydroxybutytyl-CoA by hbd encoded protein in the synthesis pathway of butyl acid in Clostridium tyrobutyricum. We designed SpyCatcher-Linker 1-thl (BBa_K4408005) and SpyTag-Linker 1-hbd to link these two enzymes together through the SpyCatcher/SpyTag system. In this way, we can improve the utilization of reagents and the reaction rate of the transformation from acetyl-CoA to 3-hydroxybutytyl-CoA, enhancing the production of butyl acid.
